Highly efficient production of transfructosylating enzymes using low-cost sugarcane molasses by A. pullulans FRR 5284

Abstract Fructooligosaccharides (FOS) are a type of important prebiotics and produced by transfructosylating enzymes. In this study, sugarcane molasses was used as the substrate for production enzymes Aureobasidium pullulans FRR 5284. NaNO 3 superior nitrogen source to yeast extract A. 5284 decreasing ratio from 1:0 1:1 resulted in reduction total activity 109.8 U/mL 82.5 U/mL. The addition only 4.4 g/L into molasses-based medium containing 100 mono- di-saccharides 123.8 Scale-up enzyme process shake flasks 1 L bioreactors improved productivity 171.7 3.58 U/mL/h, 39% 108% higher than those achieved flasks, respectively. Sucrose (500 g/L) extracellular, intracellular, enzymes, with maximum yield 61%. Intracellular, different systems FOS profiles, indicating that profiles can be controlled adjusting intracellular extracellular ratios and, hence prebiotic activity.
